It happened in the centre of the town of Espelkamp, close to the city of Bielefeld, in North Rhine-Westphalia.
One of the deceased was killed inside a house and the other just outside it, officers added.
"As things stand it looks like it was a crime rather than terrorism," a police spokesperson said.
Bild newspaper said special police commandoes had been called to the scene.
Officers found the body of a 48-year-old man and a seriously wounded woman, who later died at the scene.
A 52-year-old local man was suspected to be the gunman but there was not believed to be any danger to the wider public.
